Ion Mobility Spectrometry (IMS) is a gas phase technology that separates molecules on the basis of their size and shape. IMS is an orthogonal separation technology to HPLC/UPLC. With the introduction of electrospray ionization (ESI) and other ambient ionization techniques as a means of ionizing samples before introduction into the drift tube of an IMS instrument as well as increasing the resolving power of IMS, the recently developed high performance IMS (HPIMS) is gaining acceptance in the pharmaceutical industry as a separation technology for many applications. Some advantages of HPIMS compared to HPLC/UPLC include: 1) The speed of analysis - seconds for IMS vs. minutes for HPLC/UPLC; 2) The ability of HPIMS to separate and detect compounds that do not contain a chromophore or are unstable to aqueous reverse phase HPLC conditions; 3) HPIMS is a good separation technology for various types of isomers; 4) Due to the reduced volumes of solvents used, HPIMS is a green separation technology. We will discuss and give examples of potential uses of HPIMS in the pharmaceutical industry for: 1) Cleaning verification; 2) Reaction monitoring; 3) Dissolution studies; 4) Compounds sensitive to reverse phase chromatographic conditions; 5) Separation of isomers; 6) Using ESI-HPIMS-MS for ID confirmation.
