|LC-MS has gained broad usage in the analytical chemistry field. Based on column chemistry and choice of mobile phase (MP) composition, the retention, selectivity and separation efficiency of target analytes can be optimized. |
Reversed phase column chemistry enables the partition of non-polar analytes from the aqueous mobile phase to non-polar stationary phase and elution of the analyte by correct choice of organic MP. The eluted ions are detected by mass spectrometry. Maximizing chromatographic resolution and mass spectrometry signal of different analytes depends on the nature of MP additive, ionic strength, polarity, pH, and buffer composition. Recently use of high pH MP for basic analytes showed better mass spectral signals in ESI positive mode without any suppression of ionization.
