Course Description
This course covers the basics of high performance liquid chromatography (HPLC). All of the components of the HPLC system from the pumps to the detectors will be discussed. Troubleshooting of potential or often seen problems will be shown, whether they are the fault of the LC system, the column, or the user. The modes of chromatography, related columns and mobile phases used for various applications will be discussed with emphasis on the reversed phase mode. Basic mobile phase and sample preparation, and column care (including protector columns) will be taught since these are all critical in giving optimum method reproducibility and column life. Attendees are invited to bring their problem HPLC methods or chromatograms for discussion.

Target Audience
This course is of interest to anyone who will be doing hands-on HPLC who have not used it before or are relatively new to it. The course is designed to give an understanding of the basic principles of theory and operation to allow a HPLC method to be set up and used successfully. NOTE - this is a classroom setting so NO hands-on is possible.

Course Outline
• Chromatography theory and terminology
• HPLC system components
• Modes of chromatography
• Optimizing column configurations for speed
• Bonding chemistry and the newest LC packings for fast HPLC Afternoon
• Using the reversed phase mode
• Using the hydrophilic interaction mode (HILIC)
• Care and maintenance of LC columns
• Troubleshooting separation problems • Internet and other help sources
